[摘要] 目的 探討白藜蘆醇對缺氧缺血性腦損傷新生大鼠MMP-9及TIMP-1的影響。 方法 選擇健康7日齡新生Wistar大鼠 112隻,分為三組:假手術組(n=36隻)、缺氧組(n=38隻)、HIBD+白藜蘆醇藥物幹預組(n=38隻),各組再分為6 h、24 h、48 h、72 h、5 d和7 d時間段進行檢測。比較各組不同時間點 MMP-9、TIMP-1的表達。 結果HIBD組的MMP-9水平治療6 h、24 h、48 h、72 h、5 d、7 d分別顯著高於假手術組,但經RES幹預後,HIBD+RES組的MMP-9水平分別顯著低於HIBD組(P

[Abstract] Objective To investigate the impact of resveratrol on MMP-9 and TIMP-1 for hypoxic ischemic brain damage in neonatal rats. Methods Selected healthy 7 day old newborn Wistar rats 112 rats,were divided into three groups: sham operation group (n=36), hypoxia group (n=38 only), HIBD+resveratrol drug intervention group (n=38), each group was tested at 6 h, 24 h, 48 h, 72 h, 5 d and 7 d. The expression of TIMP-1 and MMP-9 Groups were compared at different time points. Results The MMP-9 level of 6 h, 24 h, 48 h,72 h, 5 d, 7 d in HIBD group, were significantly higher than those in the sham operation group,and was significantly higher than that of RES, but the MMP-9 levels in intervention group HIBD+RES were significantly lower than those in HIBD group(P

新生兒缺氧缺血性腦損傷(HIBD)是導致小兒智能低下和腦性癱瘓的重要原因。腦水腫是其最常見的病理改變,包括早期細胞毒性水腫和血腦屏障(BBB)損傷引起的血管源性腦水腫(VBE)。近年來研究證實,基質金屬蛋白酶-9(MMP-9)與急性期BBB的損傷密切相關,進而導致VBE的形成,而MMP-9的活性又受其金屬蛋白酶組織抑製因子(TIMP-1)的特異性抑製[1]。近年來研究發現,通過藥物抑製MMP-9在HIBD中的表達以減輕腦水腫,可能為治療缺氧缺血性腦病拓展新思路。白藜蘆醇(resveratrol,RES)是來源於植物中的一種脂溶性天然植物抗毒素,研究證實,其具有抗血小板聚集、抗炎、清除自由基、抗癌、抗氧化、心血管保護等作用[2,3]。本研究旨在探討白藜蘆醇對缺氧缺血性腦損傷新生大鼠MMP-9及TIMP-1的影響,從而為中樞神經係統的保護機製研究提供進一步的參考依據。
