麗江獐牙菜化學成分及抗乙肝病毒活性研究

[摘要] 利用各種色譜方法對麗江獐牙菜Swertia delavayi全草的化學成分進行提取、分離和純化,得到15個單體化合物,通過波譜數據分析(MS,1H,13C-NMR) 對其結構進行鑒定,分別為紅白金花內酯(erythrocentaurin, 1)、紅白金花內酯縮二甲醇(erythrocentaurin dimethylacetal, 2)、獐牙菜苷(sweroside, 3)、獐牙菜苦苷(swertiamarin, 4)、龍膽苦苷(gentiopicroside, 5)、貴州獐牙菜苷A(swertiakoside A, 6)、2′-O-乙酰基獐牙菜(2′-O-acetylswertiamarin, 7)、4′-O-[(Z)-對香豆酰基]獐牙菜苦苷(4′-O-[(Z)-coumaroyl]- swertiamarin, 8)、1,5,8-三羥基-3-甲氧基酮(1,5,8-trihydroxy-3-methoxyxanthone, 9)、8-O-β-D-吡喃葡萄糖-1-羥基-2,3,5-三甲基酮(8-O-β-D-glucopyranosyl-1- hydroxy-2,3,5-trimethoxyxanthone, 10)、8-O-[β-D-吡喃木糖-(1→6)-β-D-吡喃葡萄糖]-1羥基-2,3,5-三甲氧基酮(8-O-[β-D-xylopyranosyl-(1→6)-β-D-glucopyranosyl]-1-hydroxy-2,3,5-methoxyxant-hone, 11)、異牡荊素(isovitexin, 12)、齊墩果酸(oleanolic acid, 13)、β-穀甾醇(β-sitosterol, 14) 和胡蘿卜苷(daucosterol, 15),其中化合物1~4, 7~11, 13是首次從麗江獐牙菜中分離得到。在體外利用HepG 2.2.15細胞係對得到的單體化合物進行了抗乙肝病毒活性測試,其中化合物1,2,6,7,9和12具有明顯的抑製HBV DNV的複製活性,其IC50值為0.05~1.46 mmol·L-1。
